General description

TMB底物是辣根过氧化物酶-HRP的显色底物。

Application

TMB底物可用于利用辣根过氧化物酶结合物进行的ELISA程序。TMB底物会形成可溶的蓝色反应产物,可通过370或655 nm处的吸光度测定。该试剂可与样品中的亚硝酸盐反应,形成紫色的偶氮产物,可在450 nm处测量其吸光度。该反应可利用酸终止,形成黄色反应产物,并可通过450 nm处的光强精确测定。

我们的TMB底物使用方法:
  • 我们的TMB底物以即用型过氧化物酶底物的形式提供,其中的TMB溶于弱酸性缓冲液中。与过氧化物酶反应之前,底物溶液应呈无色至浅蓝绿色。与过氧化物酶反应时,底物系统形成蓝色反应产物。
  • 加入酸终止反应后,测量TMB底物在450 nm处的光强。
  • 我们的TMB底物推荐用于ELISA(微孔)程序,且已应用在各种测定方案中,如作为辣根过氧化物酶抗体偶联物的底物,用于检测亨德拉病毒糖蛋白、十四糖、sIgA和嗜中性白细胞胞外阱(NETs)。
  • 不建议采用3,3′,5,5′-四甲基联苯胺(TMB)液体底物系统用于膜应用。由于TMB底物会产生可溶性反应产物,因此不建议用于组织化学或印迹分析。
注意:TMB底物为光敏材料,应避免接触直射阳光或紫外线。
