General description

Lys9上乙酰化的单克隆抗-乙酰基组蛋白H3（Ac-Lys9）可识别人组蛋白H3。乙酰化的组蛋白H3免疫肽可特异性抑制免疫印迹中的组蛋白H3-Ac-Lys9条带的染色，而未乙酰化的组蛋白H3免疫肽则不会发生抑制。

Immunogen

合成的乙酰化组蛋白H3肽（氨基酸7-20，Ac-Lys9）对应于人类组蛋白H3的N端。该序列在许多物种中都相同，包括小鼠、大鼠、牛、鸡、青蛙、果蝇和 线虫，并且在 四膜虫 组蛋白H3中高度保守（单个氨基酸置换）。

Application

Biochem/physiol Actions

乙酰基组蛋白H3 组蛋白乙酰转移酶 (HAT)（包括 Gcn5p、PCAF、p300/CBP和 TAFII250）引起的组蛋白N末端结构域中赖氨酸残基的乙酰化作用与转录激活有关。这种修饰使核小体结构重塑成为一种让转录复合体更容易接近的开放式构象。反之，组蛋白脱乙酰酶（HDAC）引起的组蛋白脱乙酰作用与逆转染色质重塑过程的转录抑制有关。在大多数物种中，组蛋白H3主要在赖氨酸9、14、18和23乙酰化。赖氨酸 9上的乙酰化作用似乎在一些生物的组蛋白沉积和染色质组装中起主要作用。 组蛋白H3中特定赖氨酸的乙酰化作用也与转录之外的过程有关。在DNA复制过程中，新组蛋白被迅速合成并在复制的DNA上组装。预乙酰化状态的组蛋白H3和H4被用来复制染色质，在复制完成且新组装染色质成熟后，转变成去乙酰化状态。
