Synthesis in 4 Simple Steps

The platform consists of the Synple 2 chemical synthesizer and a range of reagent cartridges for different reactions. Simply add your starting material and the appropriate reagent cartridge to the synthesizer and press a button. The product is automatically generated, isolated and purified with only minutes of hands-on time and is running autonomously by itself during the reaction – not hours as in manual synthesis.

1. Scan the pre-filled reagent cartridge and insert it into the synthesizer, then add your starting material.

2. Simply press the START button on the synthesizer.

3. The synthesizer generates, isolates, and purifies your product in minutes.

4. Synple then performs an automated washing sequence to prepare for your next synthesis.


How Synple Works

Synple 2 Synthesizer

The Synple 2 synthesizer: contains two syringe pumps, and two rotary valves, in addition to a sample holder, cartridge holder, an RFID chip scanner, and a touchscreen. During synthesis, reactions can occur in the cartridge itself, or in the reaction vial.

Synple Cartridges

Synple cartridges: are offered for a variety of reactions. Each cartridge contains all the reagents required to generate, isolate, and purify your product, as well as an RFID chip which is encoded with the reaction method.

Automatic Synthesis

5-Minute setup: Scan the cartridge on the synthesizer to automatically load the appropriate reaction method. Although the provided methods are suitable for a broad range of starting materials, you may adjust certain parameters, such as temperature or reaction time. To initiate the reaction, insert the cartridge into the synthesizer, add your starting material, and press START.

Automatic synthesis: The starting material is prepared (e.g., dissolved), and the solution is then circulated between the vial and the first cartridge compartment for a pre-defined time. Next, it is circulated through a further number of compartments, depending on the reagents required for the reaction.

Automatic cleaning: After your product is generated, the synthesizer performs automated washing so it’s contamination-free and ready for your next synthesis.

Unparalleled speed: While manual chemical synthesis can take up to several hours to complete, Synple 2 just requires 5 to 10 minutes of the user’s time.

N-heterocycle formation cartridges simplify the conversion of a broad range of structurally and electronically diverse aldehydes into N-heterocycles, such as morpholines, piperazines, oxazepanes, and diazepanes, as well as complex spiro and bicyclic structures. They contain all reagents and catalysts needed for imine formation, cyclization, and purification.

What is in the Degrader Building Block Formation Cartridges?

Protein degrader formation cartridges

Protein degrader formation cartridges are designed for PROTAC® synthesis, and are available with either VHL or CRBN ligands in a variety of PEG linker lengths. They contain all reagents needed for linking the aldehyde or ketone group of the protein binder with the protein degrader building block, as well as for purification of the final product.

Mitsunobu - Cartridge contents

The Mitsunobu cartridge allows automated reactions to form carbon-carbon bonds through dehydrative coupling of a primary or secondary alcohol with a range of pronucleophiles, such as phenols, phthalimides, tosylamides, and tosylhydrazones. Unlike manual synthesis, there is no risk of by-product formation, thus PPh3O removal is not required.
