General description

ErbB-2原癌基因又称Neu、EGFR-2或HER-2，是跨膜受体酪氨酸激酶家族的成员，该家族还包括EGF受体和EGFR-3（HER-3或ErbB-3）。虽然ErbB-2没有已知的高亲和力配体，但是它的激酶活性可以在没有配体的情况下通过过表达或与ErbB家族其他成员的异源结合而被激活。在近40%的原发性人类乳腺肿瘤中，ErbB-2基因扩增及其产物的过度表达与淋巴结阳性乳腺癌预后不良有关。ErbB-2在卵巢癌、胃癌、涎腺癌和非小细胞肺癌中也有过表达。因此，ErbB-2是乳腺癌和其他癌症治疗的主要药物靶点之一。
