Application
免疫印迹：1-5 μg/mL，由于BDNF不容易在标准缓冲液中提取，我们建议用Semba-Katoh, R等人（J. Neurochem. 69(1):34- 42,1997）的方法制备用于蛋白质印迹的BDNF样品。切开的组织应在10倍体积的100mM磷酸盐缓冲液中匀浆，该缓冲液包含1mM EDTA，2M盐酸胍（pH7.2）和三种蛋白酶抑制剂，10mM N-乙基马来酰亚胺，0.36 mM胃抑素和1mM PMSF。然后对匀浆进行超声处理，并在4°C下以46K x g离心30分钟。 BDNF以~12-18kDa的单体迁移，在大多数样品中具有~12-18kDa二聚体。
