General description

纤连蛋白（FN）是细胞外基质（ECM）和体液中的高分子量粘着糖蛋白。它是由两个二硫键连接的两个亚基组成的二聚体。FN的每个单体由三个重复单元组成：12个I 型FN重复单元（每个单元含40个左右氨基酸）、两个II型重复单元（大约60个氨基酸）和15-17个III型重复单元（大约90个氨基酸）。这些重复单元有助于形成纤连蛋白分子结构和功能的多样性。

Application

人纤连蛋白（HFN）当用于涂覆细胞培养表面（包括塑料制品、玻璃器皿和微载体珠子）时，适合用作体外细胞繁殖的附着因子。

Biochem/physiol Actions

纤连蛋白在各种细胞相互作用的介导中起关键作用，并参与各种过程，包括细胞粘附、细胞形态的建立和维持、细胞迁移、生长和分化。它与多种细胞外基质（ECM）和细胞表面蛋白（如胶原蛋白）、肝素、血纤维蛋白和特定细胞膜受体相互作用。纤连蛋白也可作为多种整联蛋白的的配体，包括总所周知的纤连蛋白受体 α5β1，该受体结合纤连蛋白III-10 重复区域中的精氨酰-甘氨酰-天冬氨酸序列。这种广泛的相互作用网络突显了纤连蛋白在调节细胞行为和介导组织功能中的重要性。

Physical form

50 mM tris 缓冲盐水 (TBS) 溶液，pH 7.5，不含防腐剂。从 50 mM tris 缓冲盐水 (TBS) 冻干，pH 7.5，不含防腐剂（100 MG 包装规格）。37°C下使用无菌平衡盐溶液将整个 hFN 小瓶重悬为 1 mg/mL。

Analysis Note

220 kDa 的双带在还原条件下存在。hFN 是通过在明胶琼脂糖上的亲和层析，然后在肝素-琼脂糖上层析进行纯化的。已筛选出来自供体的血浆，并显示对 HIV、HTLV、乙型肝炎和丙型肝炎呈阴性。

Other Notes

涂覆细胞培养皿的建议操作步骤

1.将总表面积（cm2）乘以目标HFN 浓度（μg/mL），确定覆盖培养皿所需的 HFN 量。建议用量为 2-10 μg/cm2.。

2.采用可涂覆整个培养皿的最少量的无菌平衡盐溶液（无血清和蛋白），润湿每个培养皿的表面。

3.如果需要，通入合适的CO2气氛。

4.将计算出的 HFN 量添加到每个培养皿中。

5.让 HFN 吸附到容器表面 5-20 分钟。

6.在进行标准细胞培养程序之前，去除残留的平衡盐溶液


在生产过程中，产品使用 0.1 微米过滤器进行过滤。建议在使用前/重悬后再次过滤产品。
