M1生物测定

1.使用体外半固体琼脂培养物进行M1生物测定，该培养物在1 mL体积的DME（含20%FCS的0.3%琼脂）中含有约100个细胞。

2.加入100 μL样品或mLIF（等渗盐水中5%FCS中的10（E4）单位/mL），一式两份两倍系列稀释至35 mm培养皿。

3.将100μ L 5%FCS的等渗盐水加入到两个对照载玻片上。

4.在37°C、含10%CO2的完全湿润空气中培养7天。

5.对显示分化的菌落数进行评分（注：50单位定义为导致50%菌落分化的活性量）。
