General description
NOTCH3是一种神经元细胞命运调节蛋白，最初充当膜结合配体Jagged1、Jagged2和Delta1的受体。NOTCH3基因功能通过NOTCH3的细胞内结构域(NICD)起作用，其与RBPJ/RBPSUH形成转录激活复合物并激活分裂基因座增强子的基因，其最终影响细胞分化、增殖和凋亡程序。NOTCH3在胎儿和成人组织中广泛表达。NOTCH3蛋白在内质网中以无活性形式开始，然后在质膜中加工成活性形式。在功能上，NOTCH3被TACE裂解产生称为NEXT的膜结合细胞外片段，然后被γ-分泌酶裂解以从膜释放NICD细胞内肽，然后继续转移至细胞核以进行基因激活。NOTCH3的突变会导致脑动脉病，伴有皮层下梗塞和白质脑病综合征以及肌纤维瘤病婴儿2型疾病，这两种疾病都严重使遗传疾病衰弱。致病突变影响NOTCH3（N3ECD）外胚层结构域中表皮生长因子样重复结构域内的半胱氨酸残基。伴有皮层下梗塞和白质脑病(CADASIL)的脑常染色体显性遗传性动脉病的主要生化和组织病理学标志之一是N3ECD在血管平滑肌细胞的细胞表面积聚，并在疾病过程中退化。
