胚胎干（ES）细胞是来自胚胎植入前哺乳动物胚胎内部细胞团（ICM）的全能细胞，能够在体外无限、未分化增殖[1,2,3]。未分化的鼠类ES细胞可以在含有细胞因子、白血病抑制因子（LIF）或Millipore专有的ES细胞培养试剂ESGRO的培养基中实现长时间的体外维持′ [4,5]。 ES细胞的未分化状态表现为碱性磷酸酶（AP）的高水平表达[6]、包括SSEA和TRA抗原的表面标志物以及转录因子Oct-4的表达。

Millipore′碱性磷酸酶检测试剂盒（货号SCR004）是一种特异性和灵敏的工具，可通过确定AP活性来对ES细胞分化进行表型评估。

可从Millipore单独购买单克隆抗体TRA-2-49（产品货号MAB4349）和TRA-2-54（产品货号MAB4354），其可用于检测ES细胞表达的肝/骨/肾AP同工酶。当用于流式细胞术时，这些试剂可以定量评估ES细胞对AP表达的影响[7]。

Preparation Note

碱性磷酸酶检测试剂盒由用于确定AP活性的两个组分组成。

当存储在2-8º C，试剂盒的组分可在有效期内一直保持稳定。请勿冷冻或暴露在高温下。到期后，请丢弃所有剩余的试剂。
