General description

辣根过氧化物酶标记的抗荧光素多克隆抗体的Fab片段。该试剂是辣根过氧化物酶(POD)标记的绵羊抗荧光素抗体Fab片段。用抗荧光素免疫后,采用离子交换色谱法纯化绵羊IgG(免疫球蛋白G),然后采用免疫吸附法分离特异性IgG。采用凝胶过滤法对通过木瓜蛋白酶消化得到的Fab片段进行纯化,将其与POD偶联,然后稳定在含0.4%牛免疫球蛋白(w/v)和0.2% Germall II(w/v)、pH 7.2的60 mM Tris-Hepes缓冲液中。

Biochem/physiol Actions

该多克隆抗体可与游离和结合的荧光素反应。

Preparation Note

工作浓度:结合物的工作浓度取决于应用类型和底物。以下浓度可作为指导:
  • 斑点印迹:150 mU/ml
  • ELISA: 50至150 mU/ml
  • 免疫组织化学:250至500 mU/ml
  • 原位杂交:1.5至7.5 mU/ml
  • Southern blot:150 mU/ml
  • Western blot:500至1000mU/ml

工作溶液: 100 mM Tris-HCl,150 mM NaCl,pH 7.5必要时可使用1%封闭试剂 (w/v)、干奶粉、1-5%热灭活胎牛血清(v/v)或绵羊正常血清,以减少非特异性结合。
加入1ml双蒸馏水,最终浓度为150 U/ml。
