General description

Folin & Ciocalteu酚试剂最常用于Lowry法测定蛋白浓度。它还用于总酚类的定量。在该方法中,蛋白质用改进的缩二脲试剂(用酒石酸钠钾稳定的碱性铜溶液
)中的铜(II)进行预处理。酚试剂的加入可产生在550-750 nm间提高吸光值的色原。通常,在尖峰(750 nm)或肩峰(660 nm)处的吸光度用于定量1-100 mg/ml之间的蛋白浓度,而550nm处的吸光度用于定量更高的蛋白浓度。
在没有铜的情况下,颜色强度主要由蛋白中的酪氨酸和色氨酸含量决定,并在较小程度上由半胱氨酸和组氨酸决定。铜(II)对由酪氨酸、色氨酸或组氨酸而形成的颜色没有影响,但会由于半胱氨酸而减少颜色形成。
