General description

胶原是包含三重螺旋结构的细胞外基质蛋白。胶原蛋白I型结构特有3个相同的α-链组成同源三聚体形式。这3条α-链中有一条具有轻微的序列变化。I型胶原最初分离自鼠尾肌腱，具有两条主α链（α和α）和一条β链。

Application

大鼠尾来源I型胶原溶液已用于包被人类肝细胞HepG2细胞系和足细胞的培养板和培养皿。此外，它还还被用作绘制总胶原含量校准曲线的标准品，以定量肝细胞外基质制胶。

用作涂层材料用于支持贴壁细胞的生长和分化。

Biochem/physiol Actions

胶原蛋白是维持肌腱和骨骼机械完整性的必要成分。大鼠尾腱胶原可用于各种组织工程应用，尤其是制备基于3D支架的凝胶。这种成分具有低抗原性，同时兼容人牙龈成纤维细胞和人口腔角质形成细胞。

SDS聚丙烯酰胺凝胶电泳分析显示I型胶原蛋白的典型条带模式是具有表观分子量为115和130kDa的双峰，以及215和235kDa的另一个双峰。基于该分析可知胶原蛋白的纯度为>95%。

Other Notes

当C3867刚从冷藏箱中取出时，它呈凝胶状。完全达到37°C后，它呈液态。

胶原蛋白被分为许多种结构和遗传性不同的类型。我们使用Bornstein和Traub提出的命名法。不要混淆Sigma分类标识和公认的胶原蛋白分类类型。
