Biochem/physiol Actions

(S)-(+)-喜树碱和DNA拓扑异构酶I复合体不可逆地结合，抑制拓扑异构酶I剪切后DNA重组，将酶和DNA以共价键形式锁住。酶复合体被泛素化，被26S蛋白酶破坏，耗尽细胞内拓扑异构酶I。该酶在低剂量下在S期阻碍细胞周期，在大多数正常和肿瘤细胞中通过依赖细胞周期和不依赖细胞周期的过程诱发细胞凋亡。
