General description

β-微管蛋白属于微管蛋白亚家族。它有两个由α-螺旋包围的β-折叠。它还含有一个具有鸟嘌呤核苷酸结合区的N-末端结构域，一个具有紫杉醇结合位点的中间结构域，以及一个含有分子马达蛋白结合表面的C-末端结构域。人β-管蛋白由七种亚型组成（β（I 类）、βII（II 类）、βIII（III 类）、βIVa（IVa 类）、βIVb（IVb 类）、βV（V 类）和βVI（VI 类） ）。TUBB1（微管蛋白β I 类）基因位于人类染色体 6p21.33 上。(10)
微管蛋白是微管的主要成分。这种细胞内圆柱形丝状结构存在于几乎所有真核细胞中。微管在有丝分裂、细胞内转运、鞭毛运动和细胞骨架中充当结构和移动元件。微管蛋白是一种异二聚体，由α-微管蛋白和β-微管蛋白组成。这两个亚基的分子量均为55kDa，并具有可观的同源性。 目前研究最广泛的微管蛋白分离自脊椎动物大脑。微管可以在免疫荧光显微镜下观察，可观察超分子结构形式的蛋白质的细胞内组织。

Application

FITC-β微管蛋白抗体可用于检测β-微管蛋白的定位，通过直接标记从人胚肾细胞（HEK293T）到红豆上胚轴细胞的各种真核细胞。 利用免疫组织化学和免疫细胞化学方案进行检测。
抗β-微管蛋白-FITC单克隆抗体可用于：
  • 评估减数分裂纺锤体的状态和染色体排列β
  • 免疫荧光研究
  • 免疫荧光显微镜观察
  • 纺锤体和染色体图像(6)

Biochem/physiol Actions

β-微管蛋白基因突变可导致各种神经元迁移障碍，如无脑回、巨脑回和多小脑回畸形。β-1微管蛋白也称为VI类或TUBB1，在血小板生成中具有至关重要的作用，被认为是免疫性血小板减少症（ITP）的潜在候选生物标志物。
该抗体可识别 β 的所有 5 种亚型-微管蛋白（β1-β5).它在免疫印迹中与β-微管蛋白的羧基末端部分中的β-Lc和β-Sc片段反应。该抗体可用于定位培养细胞或组织切片中的 β-微管蛋白。

Physical form

溶于0.01M磷酸盐缓冲盐水，pH 7.4，含有1%牛血清白蛋白以及15 mM叠氮化钠。
