General description

离子霉素钙盐是由密团链霉菌产生的一种聚醚抗生素。可将Ca2+和二价阳离子从水相提取到有机相。离子霉素通过激活钙库调控型阳离子进入通路(store-regulated cation entry)来增加Ca2+内流，并非直接作用于质膜。可诱导细胞凋亡，据认为机制是通过激活潜在的钙反应性酶。离子霉素钙盐由于是高效的Ca2+载体，十分适合研究Ca2+的跨生物膜运输。离子霉素可用于平衡细胞内外的钙离子浓度，适合用于荧光指示剂的原位校准。

另外，它是针对革兰氏阳性菌的窄谱抗生素，由密团链霉菌(Streptomyces conglobatus)的发酵液纯化而来。因其优异的300nm处最大紫外吸收和对钙离子的高亲和力，在已知的聚醚抗生素中具有独特性。

Application

离子霉素钙盐（来自密团链霉菌）可作为培养基补充剂，用于培养：

  • 免疫评估的外周血单个核细胞(PBMC)
  • 孤雌激活的卵母细胞
  • T细胞亚群，用于胞内细胞因子检测、CFSE（羧基荧光素二醋酸盐琥珀酰亚胺酯）增殖实验和抗原特异性抑制功能分析。

Preparation Note
