Application

新霉素三硫酸盐水合物用于：

确定Netrin1编码基因（Ntn1）对耳蜗外植体中氨基糖苷类抗生素的保护作用
大肠杆菌 抗生素敏感试验
小鼠骨髓移植

Biochem/physiol Actions

新霉素三硫酸盐是一种氨基糖苷类抗生素，由链霉菌产生，至少含有85%的新霉素B。用于研究抗生素的细胞毒性副作用、某些成纤维细胞血小板衍生生长因子的应答以及提取核磷脂酰肌醇4,5-二磷酸相互作用蛋白。新霉素三硫酸盐可用作筛选剂，筛选出用新选择标记基因转化的原核细胞。本产品用于细胞培养时的建议使用浓度为5 mL/L。

作用方式：该产品通过和30S以及50S亚基结合，引起错编，抑制蛋白质合成过程的起始和延长。新霉素也可阻断电压敏感的Ca²⁺ 通道，而不会影响神经元中Na⁺/Ca²⁺逆向转运蛋白。

抗菌谱：新霉素能够作用于革兰氏阳性和阴性细菌。

Preparation Note

新霉素硫化物50mg/ml溶于H₂O，得到澄清溶液。

保持容器密封，存放在干燥，通风良好的地方。保持干燥。

储备溶液应过滤灭菌并在2-8℃储存，且在37℃可稳定储存5天。 1mg/mL新霉素溶液（溶于1M磷酸缓冲液），pH8.0，0-5℃储存时应在14天内使用。溶液要避光，避免潮湿。
