Application

Biochem/physiol Actions

与许多其他肽酶不同，胃蛋白酶只水解肽键，而不水解酰胺或酯键。其裂解特异性包括肽键两侧含有芳香酸的肽段，特别是当另一残基也是芳香或二羧酸时。如果肽键附近有含硫氨基酸，即含有芳香氨基酸，则易水解。胃蛋白酶也会优先在苯基丙氨酸和亮氨酸的羧基侧裂解，在谷氨酸残基的羧基侧裂解的程度较小。它不裂解缬氨酸，丙氨酸，或甘氨酸键。Z-L-酪氨酸、Z-L-谷氨酰胺-L-酪氨酸或Z-L-蛋氨酸-L-酪氨酸可作为胃蛋白酶消化的底物。胃蛋白酶被几种含有苯丙氨酸的肽所抑制。
优先裂解：P1 和 P1′ 位的疏水性和芳族残基。降解胰岛素 β 链中的 Phe-Val、Gln-His、Glu-Ala、Ala-Leu、Leu-Tyr、Tyr-Leu、Gly-Phe、Phe-Phe 和 Phe-Tyr 键

Analysis Note

最适pH值为2-4。在4 M尿素和3 M盐酸胍中可保持活性。在60°C时稳定。胃蛋白酶在pH 8.0-8.5下发生不可逆灭活。
