General description

抗-层粘连蛋白单克隆抗体（小鼠IgG1同种型）来自于小鼠骨髓瘤细胞和用人胎盘纯化层粘连蛋白免疫的小鼠脾细胞融合而成的LAM-89骨髓瘤。抗层粘连蛋白单克隆抗体特异性识别来自人、猫和猪的层粘连蛋白。该抗体可与层粘连蛋白反应，特异性染色血管、上皮、神经和肌纤维的基底膜。层粘连蛋白单抗不与IV型胶原、纤连蛋白、玻连蛋白和A、B、C型硫酸软骨素反应。层粘连蛋白是复杂的胞外糖蛋白，是基底膜(basement membrane)的主要成分。层粘连蛋白是由α (~400kDa)、β (~200kDa)和γ (~200kDa)三条链通过二硫键连接而成的异源三聚体(~850kDa)。

Application

Biochem/physiol Actions

层粘连蛋白有助于细胞外基质（ECM）的结构。它还在调节多种细胞过程中发挥着重要作用，例如细胞增殖、分化、粘附、迁移、细胞成形、表型稳定性和抗凋亡的速率。层粘连蛋白聚合物通过硫酸乙酰肝素蛋白聚糖和巢蛋白（巢蛋白）与IV型胶原蛋白聚合物形成超分子网络，并在基底膜（BM）稳定性中发挥重要作用。在小鼠中，该基因的突变与肌营养不良症的发生有关。

Physical form

0.01 M 磷酸盐缓冲液 (pH 7.4) 中的溶液，含 15 mM 叠氮化钠。

Preparation Note

–20°C 下储存。如需连续使用，请在 2–8°C 下储存长达一个月。对于长期储存，冷冻工作等分试样。不推荐反复冻融。如果延长储存时间后出现轻微浑浊，则在使用前离心澄清溶液。如果未在 12 小时内使用，工作稀释样品应丢弃。
