Calcium channel, L type α-1 polypeptide isoform 2 (CACNA1D) is crucial for the functioning of the brain and in hearing. It gets activated at a fast rate and withstands high inward Ca²⁺ currents even at threshold voltages. CACNA1D aids in dendritic signals linked to mitochondrial stress in the brain and is important for estrogen-stimulated Ca²⁺ influx. Studies have shown that in atrial myocytes, the C terminus of the channel translocates to the nucleus and regulates Ca²⁺-activated K⁺ channels. CACNA1D was shown to be upregulated in endometrial cancer and its knockdown showed a reduction in cell proliferation in vitro. Mutations in the gene encoding it has been associated with aldosterone-producing adenomas (APAs), where it leads to the formation of small adenomas.
