Serine/threonine-protein phosphatase PP1-alpha catalytic subunit (UniProt: P62136; also known as EC:3.1.3.16, PP-1A) is encoded by the PPP1CA (also known as PPP1A) gene (Gene ID: 5499) in human. PPP1A is one of the three catalytic subunits of protein phosphatase 1 (PP1), a major eukaryotic serine/threonine phosphatase that is involved in a multitude of cellular functions. In addition to PP1 alpha, there is PP1 beta (PPP1CB), and two splice variants of PP1 gamma (PPP1CC). PPP1A interacts with over 200 regulatory proteins to form highly specific holoenzymes which dephosphorylate a huge variety of target proteins. PPP1A is critical for cell division, chromatin structure, protein synthesis, glycogen metabolism, muscle and cardiac function as well as the regulation of long-term ionic conductances and even synaptic plasticity. Developmentally, it has nearly as many roles with principal ones being as a regulator of neural tube and optic fissure closure as well as enteric neural crest cell (ENCCs) migration during development. PPP1A protein is ubiquitously expressed in human tissues, with particularly high levels in the brain, heart, and skeletal muscle, reflecting its fundamental role in cellular signaling pathways. PPP1A can be found both cytoplasmically and nuclear localized depending upon its regulatory partner but PPP1A is mostly nuclear/nucleoplasm localized. Three isoforms of PPP1A have been described that are produced by alternative splicing. Dysregulation of PPP1A has been implicated in several diseases, including cancer, where altered phosphatase activity can lead to aberrant cell growth and survival. PPP1A has been linked to arthritis, playing distinct roles in both inflammatory and degenerative forms of the disease. It is suggested to be up-regulated in synovial fluid mononuclear cells and peripheral blood mononuclear cells from patients with rheumatoid arthritis. In the brain, PPP1A also functions as a suppressor of learning and memory and, as a potential mediator of cognitive decline during aging. It has also been associated Alzheimer′s disease. Recent studies suggest crucial role of PPP1CA in regulating HCC tumorigenesis through a YAP-dependent mechanism and identify it as a promising therapeutic target for HCC intervention. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Application
Quality Control Testing
Evaluated by Western Blotting in HeLa cell lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected PPP1A in HeLa cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PPP1A in lysates from A-431cells, C6 cells, NIH/3T3 cells, human kidney tissue, and mouse kidney tissue.

Biochem/physiol Actions
Clone 1H8 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects PPP1A. It targets an epitope within 15 amino acids from the C-terminal half.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
