Proteinase-activated receptor 2 (UniProt: P55085; also known as PAR-2, Coagulation factor II receptor-like 1, G-protein coupled receptor 11, Thrombin receptor-like 1) is encoded by the F2RL1 (also known as GPR11, PAR2) gene (Gene ID: 2150) in human. PAR2 is a member of the proteinase-activated receptor subfamily. It is activated through proteolytic exposure of an occult tethered ligand by trypsin and trypsin-like proteases. Its function is mediated through the activation of several signaling pathways including phospholipase C (PLC), intracellular calcium, mitogen-activated protein kinase (MAPK), I-kappaB kinase/NF-kappaB and Rho. Also, as a part of the innate and adaptive immunity response, PAR2 acts as sensor for proteolytic enzymes generated during infection and works to promote inflammation and the inflammatory response. PAR2 is also involved in vascular and airway tone and in blood pressure regulation and many other G protein coupled receptor response pathways. PAR2 is a multi-pass membrane protein that is widely expressed in most tissues with especially high levels in pancreas, liver, kidney, small intestine, and colon. It is expressed on multiple cell types within the central nervous system (CNS), including neurons, astrocytes, and microglia. PAR2 has been implicated in acute inflammatory response and pain transmission. Dysregulation of PAR-2 has been implicated in several diseases, including asthma, inflammatory bowel disease, and certain cancers, where its overactivation contributes to chronic inflammation and tumor progression. Recent research highlights the emerging role and interactions of PAR2 in different skin cells and provides insights into its role in skin conditions such as atopic dermatitis (AD), psoriasis, vitiligo and melasma. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. KLH-conjugated linear peptide corresponding to 17 amino acids from the C-terminal cytoplasmic domain of human PAR2.
Application
Quality Control Testing
Evaluated by Western Blotting in lysate from PANC-1 cells.
Western Blotting Analysis: A 1:1000 dilution of this antibody detected PAR2 in PANC-1 cell lysate.
Tested Applications
Western Blotting Analysis: A 1:1000 dilution from a representative lot detected PAR2 in lysates from MDA-MB-231 cells and PC-3 cells.
Enzyme Immunoassay: A representative lot of this antibody detected recombinant PAR2 protein in ELISA application.

Biochem/physiol Actions
Clone 1F8 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects PAR2. It targets an epitope within 17 amino acids from the C-terminal cytoplasmic domain.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Preparation Note
300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
