General description

Neurofilament light chain (NF-L) is a critical component of the neurofilament cytoskeleton in neurons, providing structural support and maintaining neuronal integrity. As part of the larger family of neurofilament proteins, NF-L, along with neurofilament medium chain (NF-M) and neurofilament heavy chain (NF-H), contributes to the formation of intermediate filaments that are essential for the structural stability of axons. NF-L plays an essential role in axonal transport, facilitating the movement of organelles and proteins along the axon, which is vital for neuronal function and communication.

Additionally, NF-L participates in the response to injury and stress, helping neurons adapt to changing conditions. Its presence in the central nervous system is crucial for maintaining the overall health and functionality of neurons. NF-L serves as a valuable biomarker for neuronal damage and neuroinflammation. Elevated levels of NF-L in cerebrospinal fluid (CSF) and blood indicate ongoing neurodegeneration and correlate with conditions such as Alzheimer′s disease, amyotrophic lateral sclerosis (ALS), and multiple sclerosis (MS). Additionally, increased NF-L levels often reflect the severity of neuroinflammatory processes. The measurement of NF-L is essential for developing therapeutic strategies aimed at preserving neuronal health and function in various neurological disorders.

Analysis Note

  • Sensitivity: LLOQ 870 fg/mL* (SMCxPRO® Instrument)
  • Standard Curve Range: 430-1,500,000 fg/mL
  • Intra-assay CV: <15%
  • Inter-assay CV: <20%
  • Linearity: Serum: 110% Plasma: 106% CSF: 101%
  • Recovery: Serum: 93% Plasma: 94% CSF: 123%
*Indicative LLOQ created from average of 14 independent in house runs, where >80% met same value defined with CV & Recovery +/- 20%. Refer to CoA for lot specific performance.

Other Notes

Although NF-L was detectable in mouse, non-human primate (rhesus macaque and cynomolgus monkey), and canine samples, the assay has not been verified for these species.
