Gene Information
human ... EEF1A1(1915)
General description
53 kDa
Human eukaryotic translation elongation factor 1 alpha has two isoforms, alpha 1 and alpha 2. The alpha 1 isoform gene encodes a subunit of the elongation factor-1 complex which is responsible for the enzymatic delivery of aminoacyl tRNAs to the ribosome. The alpha 1 isoform is expressed in brain, placenta, lung, liver, kidney, and pancreas. The other isoform (alpha 2) is expressed in brain, heart and skeletal muscle.
The alpha 1 isoform is identified as an autoantigen in 66% of patients with Felty syndrome. The gene for the alpha 1 isoform has been found to have multiple copies on many chromosomes, some of which, if not all, represent different pseudogenes. The gene for the alpha 2 may be critical in the development of ovarian cancer.

Biochem/physiol Actions
This antibody recognizes EF-1α (Elongation factor-1 alpha), Mr 53 kDa. Occasionally, a 38 kDa protein is also detected in A431 cell lysate.
Physical form
Format: Purified
Protein G Chromatography
Purified mouse monoclonal IgG1κ in buffer containing 0.1 M Tris-glycine, pH 7.4, 0.15 M NaCl with 0.05% sodium azide.
Preparation Note
Stable for 1 year at -20°C from date of receipt.
Handling Recommendations:
Upon receipt, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20ºC. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Control
Positive Antigen Control: Catalog #12-301, non-stimulated A431 cell lysate. Add 2.5µL of 2-mercaptoethanol/100µL of lysate and boil for 5 minutes to reduce the preparation. Load 20µg of reduced lysate per lane for minigels.

Routinely evaluated on RIPA lysates from non-stimulated A431 cells, human Jurkat or mouse 3T3/A31.
Western Blot Analysis:
0.5-2 µg/mL of this lot detected EF1α in RIPA lysates from non-stimulated A431 cells. Previous lots detected EF1α in human Jurkat and mouse 3T3/A31 RIPA cell lysates.

Regulation of translation of mRNAs coding for specific proteins plays an important role in controlling cell growth, differentiation, and transformation. Two proteins have been implicated in the regulation of specific mRNA translation: eukaryotic initiation factor eIF4E and ribosomal protein S6.

The following study examines the calmodulin (CaM) branch of the calcium signal pathway in the protozoan parasite Trypanosoma brucei. To accomplish this goal, a subset of cytosolic CaM-binding proteins (CaMBPs) was partially purified by a combination of DE52 and CaM-Sepharose
