General description

Acetylcholine (ACh) is a common neurotransmitter for motoneurons, preganglionic autonomic neurons, postganglionic parasympathetic neurons, a variety of brain regions and some emerging neuron-like stem cells. The metabolism of Ach is relatively simple, involving only two enzymes: choline acetyltransferase (ChAT) for synthesis and acetylcholinesterase (AChE) for degradation. Further, acetylcholine has little function in neurons other than neurotransmission and seems to be neuron specific. It seems that only cholinergic neurons have significant amounts of ChAT making anti-cholinacetyltransferase a useful specific marker.

Application

Anti-Choline Acetyltransferase (ChAT) Antibody is an antibody against Choline Acetyltransferase (ChAT) for use in IH, IH(P), IP & WB with more than 70 product citations.
Immunoprecipitation:
1:200 dilution of a previous lot immunoprecipitated 68 kDa active ChAT from brain and placenta. Also precipitates a 27 kDa protein that is co-expressed with ChAT (but lacking ChAT activity) in some cholinergic neurons of the central nervous system of Humans and possibly other mammals.

Optimal working dilutions and protocls must be determined by end user.

Biochem/physiol Actions

Cholinergic neurons in brain and central nervous systems.
Reactivity with other species has not been confirmed with IHC.

Physical form

Rabbit polyclonal antisera, liquid, no preservatives.

Analysis Note

Control
Cerebral cortex tissue.
Routinely evaluated by immunohistochemistry (paraffin) on normal cerebral cortex tissue.

Immunohistochemistry (Paraffin) Analysis:
ChAT (cat. # AB143) staining on normal cerebral cortex. Tissue treated with no antigen retrieval. The antibody was diluted to 1:100, using IHC-Select Detection with HRP-DAB. Immunoreactivity is seeing as a staining pattern on the cell body, dendrites and axons of neurons (brown).

Other Notes

Concentration: Please refer to the Certificate of Analysis for the lot-specific concentration.

The p75 low-affinity neurotrophin receptor (p75(LNTR)) appears to have various functions that include enhancing nerve growth factor (NGF)-mediated survival by increasing TrkA (high-affinity NGF receptor) efficiency, and mediating apoptosis by acting as a ligand-regulated pro-apoptotic receptor. Here, we investigated the
