General description
Protamine sulfate is a cationic polypeptide having a molecular weight of 5 kDa, isolated from salmon sperm.
Application
Protamine sulfate has been used in the transduction of
- lentiviruses into human embryonic 293T cells
- shRNA into mesenchymal stem cells/marrow stromal cells (MSC)
- lenti Sca1-OCT4 plasmid into human cord blood hematopoietic progenitor CD34+ cells
Biochem/physiol Actions
Alkaline protamine binds to negatively charged heparin. Use of protamine during cardiovascular surgery sometime triggers anaphylactic reactions.
Protamine facilitates direct transfection of microbial cells, and of human cells using cationic liposomes. Protamine effectively neutralizes the anticoagulation effects of heparin.

The neutralization of a low molecular weight heparin (LHN-1) and conventional heparin (CH) by protamine sulfate has been studied in vitro and in vivo. In vitro, the APTT activity of CH was completely neutralized in parallel with the anti-Xa activity.
