General description
脱氧核糖核酸酶Ⅰ将单链和双链 DNA 消化为带 5′磷酸盐和3′羟基末端的单核苷酸和寡核苷酸混合物。这种催化活性依赖于二价离子。在存在Mg2+的情况下,脱氧核糖核酸酶Ⅰ随机而独立地水解双链DNA的每条链。在存在Mn2+的情况下,两条链都可以被切开。
Application
来自牛胰脏的无RNA酶脱氧核糖核酸酶I溶液已被用于消化各种样品中的DNA。
在分子生物学应用中用于RNA纯化过程中去除DNA,用于制备DNA进行缺口翻译,以及通过足迹法分析DNA-蛋白质相互作用。
Other Notes
DNase I 溶于 50% (v/v) 甘油、20 mM 醋酸钠 (pH 6.5)、5 mM CaCl2 和 0.1 mM PMSF 溶液中。
使用小牛胸腺DNA作为底物时,一单位在每毫升反应混合物中每分钟可引起0.001的ΔA260。
用这种DNase I处理过的RNA不应该用于生成cDNA文库或用于RT-PCR。对于这些敏感的应用,建议使用扩展级DNase I,目录号AMP-D1。
