General description

TransPlex®,一种全转录组扩增(WTA)方法,能够在4小时内,代表性地扩增出纳克级的总RNA而没有3′-偏差。 从组织、培养细胞、福尔马林固定样品或血清中所获得的微克级扩增产物适用于例如qPCR和微阵列分析等下游应用。

Application

Biochem/physiol Actions

WTA过程包括2个步骤。首先,使用由拟随机3′端和通用5′端组成的非自身互补引物对样品RNA进行逆转录。随着聚合作用的发生,被取代的单链将作为引物退火和延伸的新模板。所得OmniPlex cDNA文库由以通用末端序列为侧翼序列、含100-1000个碱基的随机重叠片段组成。随后,使用通用引物对文库进行PCR扩增,生成WTA产物。
